چکیده

The role of shikimic acid as a common intermediate in the synthesis of phenylalanine, tyrosine, tryptophan, and p-aminobenzoic acid has been recognized for some time (l-3). The enzymes which catalyze the synthesis of shikimic acid from intermediates of carbohydrate metabolism have been observed in extracts of bacteria (4-6) as well as Neurospora crassa (7). On the other hand, the reactions leading from shikimic acid to specific intermediates in the synthesis of the various aromatic amino acids have been poorly understood. Only recently, studies with extracts from bacterial mutants (S-13) have implicated the sequence of reactions presented in Fig. 1. From the bacterial studies, it appears that shikimic acid is converted by three enzymatic reactions to chorismic acid, which serves as a direct precursor of the various specific pathways of aromatic biosynthesis. Genetic and nutritional studies with N. crassa indicate that the first enzyme specifically involved in tryptophan biosynthesis catalyzes the formation of anthranilic acid (14). As an initial approach toward the investigation of this enzyme in N. crassa, attempts were made to show the conversion of shikimic acid to anthranilic acid. Under the conditions employed in the bacterial studies, crude extracts of N. crassa did not catalyze this conversion. The studies presented here were undertaken to define the conditions necessary for the synthesis of anthranilic acid from shikimic acid in N. crassa and to investigate the reactions involved.

Synthesis of Aromatic Compounds by Neurospora.

Increasing insight into the biosynthesis of aromatic substances has been obtained during the past few years primarily through the study of mutant strains of microorganisms,1 and by the use of isotopic carbon.2 Shikimic acid, reported to be involved in the biosynthesis of the aromatic amino acids and of p-aminobenzoic acid by Neurospora3 and Escherichia coli, has been shown to be derived in E. c...

Incorporation of carbon atoms 1 and 6 of glucose into protocatechuic acid by Neurospora.

The mechanism of biosynthesis of the simpler aromatic compounds has been significantly clarified recently by the use of mutant strains of Neurospora (1) and Escherichia coli (2) and by the use of isotopic carbon (3-6). The evidence indicates that aromatization takes place via dehydroquinic acid, dehydroshikimic acid, and shikimic acid. Although the ultimate source of the carbon atoms of the aro...

The metabolism of protocatechuic acid by Neurospora.

A mutant strain of Neurospora crassa, Y7655a, which requires for growth either phenylalanine, tyrosine, tryptophan, and p-aminobenzoic acid in combination or shikimic acid alone, was found to be blocked in the conversion of dehydroshikimic acid t.o shikimic acid. Some dehydroshikimic acid was accumulated during growth, but the major accumulated product was protocat.echuic acid (3,4-dihydroxyben...

The enzymatic conversion of 5-dehydroshikimic acid to protocatechuic acid.

Previous studies of the physiology of an aromatic-deficient mutant strain of Neurospora crassa, Y7655a (1, 2), have led to the following conclusions: (a) The biosynthesis of the aromatic rings of phenylalanine, tyrosine, tryptophan and p-aminobenzoic acid proceeds in Neurospora as it does in Escherichia coli (3) via 5-dehydroshikimic acid and shikimic acid. (b) Protocatechuic acid, the major co...

Alteration of tryptophan-mediated regulation in Neurospora crassa by indoleglycerol phosphate.

The accumulation of imidazoleglycerol phosphate during growth of Neurospora crassa in the presence of 3-amino-1,2,4-triazole was found to cause derepression of tryptophan synthetase and to inhibit the induction of kynureninase. Accumulation of indoleglycerol phosphate in response to growth in the presence of indole acrylic acid or anthranilic acid was also accompanied by derepressed synthesis o...
